Small bore (120 µm ID) Teflon or PEEK tubing is used to reduce the delay between dialysis and sample collection. The swept volume is ca. 1.2 µL/10 cm. Tubing is sold in 1 m lengths and is connected to 23 ga. syringe needles, dialysis probes, swivels, and cannulae (0.6 - 0.8 mm OD) by means of a flanged plastic connector. The connector "splices" the tubing and cannula together, eliminating dead space between them.
If bacterial contamination is a concern between tubing uses (and it should be if you are working with ACh, amino acids, glucose, lactate, or pyruvate in particular), flush some diluted ProClin150 (CF-2150) through the tubing between uses. [Note: if a liquid swivel is plumbed in your system, do NOT use a syringe to manually fill the tubing and/or liquid swivel with the ProClin. If you do, you will likely damage your swivel. Instead, use a syringe pump to flush the tubing and swivel with the ProClin at normal microdialysis flow rates, or aspirate the ProClin solution through the system via suction from a syringe.] Flush several volumes of water or perfusate though the system before reusing tubing or swivel flushed with ProClin150.
