The use of enzymes as selective reagents in liquid chromatography was first reported in the early 1970's. For the neurochemicals acetytlcholine (ACh) and choline (Ch), enzymes provide an essential means for their detection. The compounds are neither electroactive nor UV-absorbing under reasonable conditions. However, via enzyme catalyzed reactions both compounds are converted to hydrogen peroxide, a compound that may be either oxidized or reduced.

The enzymes acetylcholinesterase and choline oxidase are covalently attached to a solid matrix and packed into a column. The result is an immobilized enzyme reactor (IMER).

First in line, the analytical column separates ACh and Ch as a pair well-resolved from unretained peaks. The second column, a ready-to-use IMER, is coupled to the outlet of the analytical column and catalyzes the production of hydrogen peroxide in direct proportion to the concentration of ACh and Ch. The peroxide is detected by oxidation on a platinum working electrode or by reduction on a modified glassy carbon wired electrode.
Two versions of the kit are available for ACh and Ch determination: a cartridge-column version and a UniJet microbore version. In general, the microbore version should be used where ACh concentration and sample volume are low, such as samples from in vivo microdialysis.
The Kits are shipped via next-day service. They are packed in special cold-pack containers to preserve enzyme activity. Upon receipt, the kits should be refrigerated (NOT FROZEN) until used. An antibacterial is included in the kits to inhibit the growth of bacteria in the chromatography system and columns.

The UniJet Microbore Acetylcholine/Choline kit was developed to provide improved sensitivity, especially with in vivo microdialysis samples. The kit includes a capillary flow splitter, microbore analytical and IMER columns, standards, and other necessary accessories. Requires a BASi electrochemical detector, dual platinum or wired working electrode, and a liquid chromatograph.
